Bioprocess development and optimization in shaking flasks with high resolution exhaust gas analysis.
RAMOS (Respiration Activity MOnitoring System) has established itself as a world wide standard tool for the online detection of the respiration activity in shaking flask with prokaryotic and eukaryotic cultures.
The increasing demand of pharmaceuticals originated in biotechnology, especially from mammalian cell cultures, makes specific optimisation indispensable. Beside the oxygen transfer rate (OTR), the carbon dioxide transfer rate (CTR) and the respiratory quotient (RQ)
are important sources of information for microbiologists and bioprocess engineers. The CTR makes the RAMOS even suitable for anaerobic processes and microaerophilic organisms. Information regarding the carbon source can be derived from the RQ, i.e. a RQ=1 is typical for glucose and RQ=0.65 for triglycerides. Depending on the users experience even complex metabolic reaction can be analysed.
The proven RAMOS® bio-process optimisation system is now also available for disposable flasks. Corning flasks offer an alternative to glass shake flasks, especially for cell culture equipment. But the RAMOS® may also be adjusted for other disposable flasks. Controlled fed-batch fermentations are also possible in disposable flasks with the proven feed module. The feed profiles may be composed of constant, linear and exponential processes.
RAMOS® records the processes of oxygen consumption and carbon dioxide production of the cultures and is thus the perfect system for bio process optimisations. Through the feed system it is possible, in addition to batch tests, to carry out fed-batch tests, which until now were restricted to laboratory fermenters, both in standard RAMOS® and disposable flasks.
Existing RAMOS® systems can be retrofitted.
